Zeytinya\u011f\u0131n\u0131n sabunla\u015fmayan k\u0131sm\u0131 fenolik bile\u015fikleri i\u00e7erirken, sabunla\u015fabilen k\u0131sm\u0131 ise oleik asit gibi tekli doymam\u0131\u015f ya\u011f asitleri (MUFA) a\u00e7\u0131s\u0131ndan zengindir [46]. Fenolik bile\u015fikler \u00fc\u00e7 gruba ayr\u0131l\u0131r: basit fenoller (\u00f6r. tirosol, hidroksitirosol veya 3,4-dihidroksifeniletanol), fenolik asitler (\u00f6r. kafeik asit) ve flavonoidler (\u00f6r. kuersetin). En \u00f6nemli kompleks fenoller aras\u0131nda tirosol ve hidroksitirosol esterleri, oleuropein ve onun aglikonu bulunmaktad\u0131r [47]. Hidroksitirosol, tirosol ve oleuropein, benzer yap\u0131lara sahip olup zeytinya\u011f\u0131nda y\u00fcksek miktarlarda bulunur [48] (Tablo 2).<\/p>\n<p>Ara\u015ft\u0131rmalar, zeytinya\u011f\u0131 polifenollerinin h\u00fccresel DNA\u2019da olu\u015fan oksidatif hasar\u0131 azaltarak CRC geli\u015fimini engelleyebilece\u011fini g\u00f6stermektedir. \u00d6zellikle hidroksitirosol ve oleuropein gibi y\u00fcksek antioksidan polifenoller, zeytinya\u011f\u0131 depolama s\u0131ras\u0131nda olu\u015fabilecek potansiyel kanserojen lipid peroksidasyon \u00fcr\u00fcnlerini azaltabilir, pozitif epigenetik de\u011fi\u015fikliklere ve miRNA ifade d\u00fczenlemelerine yol a\u00e7arak CRC riskini d\u00fc\u015f\u00fcrebilir [49\u201351]. Oksidatif stres, organizman\u0131n oksidan ve antioksidan sistemleri aras\u0131ndaki dengesizlikten kaynaklan\u0131r ve genellikle serbest radikaller ile reaktif oksijen t\u00fcrlerinin (ROS) neden oldu\u011fu oksidasyon sonucu ortaya \u00e7\u0131kar. Bu serbest radikaller, ya\u011f asitlerine zarar vererek membran fonksiyonu ve b\u00fct\u00fcnl\u00fc\u011f\u00fcn\u00fcn kayb\u0131na, ard\u0131ndan da apoptoz ve nekroza yol a\u00e7abilecek zincir reaksiyonlar\u0131 ba\u015flatabilir [52,53].<\/p>\n<p>Bir\u00e7ok \u00e7al\u0131\u015fma, hidroksitirosol\u00fcn (HT) \u00f6nemli antiinflamatuvar [54] ve antit\u00fcm\u00f6r etkileri [55] oldu\u011funu g\u00f6stermi\u015ftir. HT\u2019nin di\u011fer faydalar\u0131 aras\u0131nda endotelyal h\u00fccre fonksiyonlar\u0131n\u0131n iyile\u015ftirilmesi [56], karaci\u011fer steatozu \u00fczerinde koruyucu etkisi [57] ve n\u00f6roprotektif \u00f6zellikleri [58] yer al\u0131r. Avrupa G\u0131da G\u00fcvenli\u011fi Otoritesi (EFSA) Diyet \u00dcr\u00fcnleri, Beslenme ve Alerjiler Paneli, HT\u2019nin d\u00fc\u015f\u00fck yo\u011funluklu lipoproteinleri (LDL) oksidatif modifikasyonlara kar\u015f\u0131 koruyabilece\u011fini belirtmi\u015f ve g\u00fcnl\u00fck en az 5 mg HT ile t\u00fcrevlerinin (\u00f6r. oleuropein kompleksi ve tirosol) t\u00fcketilmesini \u00f6nermi\u015ftir [59].<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignnone wp-image-96 size-mh-magazine-lite-content\" src=\"https:\/\/zeytinkokusu.com\/blog\/wp-content\/uploads\/2025\/01\/Kisibasina-zeytin-tuketimi-dunyada-1-678x381.jpg\" alt=\"\" width=\"678\" height=\"381\" \/><\/p>\n<p>Mateos ve arkada\u015flar\u0131n\u0131n bir \u00e7al\u0131\u015fmas\u0131, zeytinya\u011f\u0131 HT asetat\u0131n\u0131n insan kolon adenokarsinoma h\u00fccreleri \u00fczerinde antit\u00fcm\u00f6r aktivite g\u00f6sterebilece\u011fini ve programlanm\u0131\u015f h\u00fccre \u00f6l\u00fcm\u00fcnde yer alan genlerin (BNIP3, BNIP3L, PDCD4 ve ATF3) transkripsiyonunu etkileyerek kaspaz-3 aktivasyonunu tetikleyebilece\u011fini ortaya koymu\u015ftur. HT asetat ayr\u0131ca, karsinojen detoksifikasyonunu art\u0131rarak UGT1A10 ve CYP1A1 gibi ksenobiyotik metabolize eden enzimleri d\u00fczenleyebilir [28]. Di\u011fer ara\u015ft\u0131rmalar, HT\u2019nin \u00e7e\u015fitli t\u00fcm\u00f6rlerde h\u00fccre d\u00f6ng\u00fcs\u00fcn\u00fc durdurup apoptozu ind\u00fckleyerek kanser \u00e7o\u011falmas\u0131n\u0131 engelleyebilece\u011fini g\u00f6stermi\u015ftir; buna kolanjiyokarsinom da dahildir [60].<\/p>\n<p>HT, epidermal b\u00fcy\u00fcme fakt\u00f6r\u00fc resept\u00f6r\u00fcn\u00fcn (EGFR) ifadesini azaltabilir ve kolon kanseri h\u00fccrelerinde h\u00fccre d\u00f6ng\u00fcs\u00fcn\u00fc inhibe ederek cetuximab gibi EGFR\u2019ye kar\u015f\u0131 monoklonal antikorlar\u0131n etkisini taklit edebilir [29]. Terzuoli ve ekibinin yak\u0131n tarihli bir \u00e7al\u0131\u015fmas\u0131, cetuximab kemoterapisi s\u0131ras\u0131nda zeytinya\u011f\u0131 i\u00e7eren kontroll\u00fc bir diyetin potansiyel faydalar\u0131n\u0131 \u00f6nermektedir [30]. HT, EGFR inhibit\u00f6rlerinin etkisini art\u0131rarak kolon kanseri hastalar\u0131nda faydal\u0131 bir terap\u00f6tik ajan olarak rol oynayabilir [30]. \u0130lgin\u00e7 bir \u015fekilde, kurkumin ve resveratrol gibi di\u011fer polifenollerin de kemoterap\u00f6tik ajanlarla kombine edildi\u011finde CRC \u00fczerinde terap\u00f6tik faydalar sa\u011flad\u0131\u011f\u0131 g\u00f6r\u00fclm\u00fc\u015ft\u00fcr [61,62].<\/p>\n<p>Zeytin karasuyu (OMWW) kaynakl\u0131 HT a\u00e7\u0131s\u0131ndan zengin safla\u015ft\u0131r\u0131lm\u0131\u015f ekstraktlar\u0131n, hem in vivo hem de in vitro olarak CT-26 CRC h\u00fccre hatt\u0131nda anti-anjiyojenik ve kemopreventif etkiler g\u00f6sterdi\u011fi bulunmu\u015ftur [13].<\/p>\n<p>Rossi ve arkada\u015flar\u0131, OMWW\u2019den elde edilen ve A009 olarak adland\u0131r\u0131lan safla\u015ft\u0131r\u0131lm\u0131\u015f, polifenol bak\u0131m\u0131ndan zengin bir ekstrakt\u0131n \u00f6zelliklerini ara\u015ft\u0131rm\u0131\u015ft\u0131r. A009, hem in vitro hem de in vivo olarak g\u00fc\u00e7l\u00fc anti-anjiyojenik etkiler sergilemi\u015ftir. Ayn\u0131 konsantrasyondaki HT\u2019nin etkisiyle kar\u015f\u0131la\u015ft\u0131r\u0131ld\u0131\u011f\u0131nda, A009\u2019un anjiyogenezi inhibe etme potansiyeli daha g\u00fc\u00e7l\u00fc bulunmu\u015ftur. Bu ekstrakt ayr\u0131ca endotelyal h\u00fccre proliferasyonu, g\u00f6\u00e7\u00fc ve invazyonunu engellemede etkili olmu\u015ftur [27]. Gen\u00e7 zeytinlerde ve zeytin yapraklar\u0131nda y\u00fcksek konsantrasyonlarda bulunur. Antioksidan, antienflamatuvar, antit\u00fcm\u00f6r, antiaterojenik ve hipoglisemik etkileri ile dikkat \u00e7eker.<\/p>\n<p>\u00c7al\u0131\u015fmalar, oleuropein&#8217;in h\u00fccre d\u00f6ng\u00fcs\u00fcn\u00fc d\u00fczenleyerek kanser h\u00fccrelerinin b\u00fcy\u00fcmesini engelledi\u011fini, apoptozu tetikledi\u011fini ve aktin filamentlerini bozarak t\u00fcm\u00f6r h\u00fccrelerinin yay\u0131lmas\u0131n\u0131 \u00f6nledi\u011fini g\u00f6stermektedir. Farelerde yap\u0131lan deneylerde, oleuropein\u2019in oral t\u00fcketiminin t\u00fcm\u00f6rlerin k\u00fc\u00e7\u00fclmesine neden oldu\u011fu g\u00f6zlemlenmi\u015ftir.<\/p>\n<p>Oleuropein\u2019in kolon kanseri \u00fczerindeki etkileri, p53 yolu ve HIF-1\u03b1 proteini gibi mekanizmalarla ili\u015fkilidir. Ayr\u0131ca inflamatuvar belirte\u00e7leri (IFN-\u03b3, TNF-\u03b2, IL-6) ve kanserle ili\u015fkili yollar\u0131 (Wnt\/\u03b2-katenin, PI3K\/Akt) bask\u0131lad\u0131\u011f\u0131 belirtilmi\u015ftir. Oleuropein\u2019in anjiyogenezi engelleyerek t\u00fcm\u00f6r b\u00fcy\u00fcmesini yava\u015flatt\u0131\u011f\u0131 ve anti-anjiyogenik ila\u00e7lara benzer \u015fekilde fayda sa\u011flad\u0131\u011f\u0131 da g\u00f6sterilmi\u015ftir.<\/p>\n<p>Oleocanthal adl\u0131 ba\u015fka bir zeytinya\u011f\u0131 bile\u015fi\u011fi, kanser h\u00fccrelerinde apoptoz ve DNA hasar\u0131na neden olarak antit\u00fcm\u00f6r etkiler g\u00f6stermektedir. Ayr\u0131ca Hsp90 gibi \u0131s\u0131 \u015fok proteinlerini inhibe ederek kanser tedavisinde yeni bir yakla\u015f\u0131m sunabilir.<\/p>\n<p>Tabii, metni \u00f6zetleyerek T\u00fcrk\u00e7ele\u015ftirdim:<\/p>\n<h3><strong>Zeytinya\u011f\u0131 Polifenolleri ve \u0130ltihap Kontrol\u00fc<\/strong><\/h3>\n<p>Zeytinya\u011f\u0131 (OO) polifenolleri, inflamatuvar tepkileri mod\u00fcle edebilir ve NF-\u03baB\u2019yi inhibe edebilir. Bu inhibit\u00f6r etki, IL-6, IL-8, IL-1\u03b2 ve COX-2 gibi inflamatuvar belirte\u00e7lerin d\u00fc\u015f\u00fck seviyelerde ifade edilmesini sa\u011flar ve kanser b\u00fcy\u00fcmesini engelleyici bir mikro\u00e7evre olu\u015fturur [73,74]. Beauchamp ve ekibi, oleocanthal\u2019in ibuprofene benzer \u015fekilde siklooksijenaz (COX) enzimlerini inhibe ederek anti-inflamatuvar etki g\u00f6sterdi\u011fini ortaya koymu\u015ftur [75]. COX inhibit\u00f6rlerinin, \u00f6rne\u011fin ibuprofen ve aspirinin, antikanserojenik ve antitrombotik etkileri iyi bilinmektedir [76,77].<\/p>\n<p>Oleocanthal\u2019in inflamatuvar ba\u011f\u0131rsak hastal\u0131klar\u0131n\u0131 (IBH) (\u00fclseratif kolit ve Crohn hastal\u0131\u011f\u0131) azaltabilece\u011fi ve dolayl\u0131 olarak kolorektal kanser (CRC) riskini d\u00fc\u015f\u00fcrebilece\u011fi d\u00fc\u015f\u00fcn\u00fclmektedir [78,79]. IBH, CRC i\u00e7in \u00f6nemli bir risk fakt\u00f6r\u00fcd\u00fcr ve CRC, IBH hastalar\u0131n\u0131n %1\u2019inde g\u00f6r\u00fclmesine ra\u011fmen y\u00fcksek mortalite oran\u0131na sahiptir [80,81,82].<\/p>\n<p>S\u0131\u00e7anlar \u00fczerinde yap\u0131lan bir \u00e7al\u0131\u015fmada, azoksimetan ile ind\u00fcklenen CRC\u2019ye kar\u015f\u0131 zeytinya\u011f\u0131n\u0131n kemopreventif etkisi g\u00f6zlemlenmi\u015ftir. Bu etkiler, zeytinya\u011f\u0131ndaki oleik asit ve eikosapentaenoik asit gibi ya\u011f asitlerinin, kolonik mukoza \u00fczerinde arakidonik asit metabolizmas\u0131n\u0131 ve prostaglandin E2 sentezini mod\u00fcle etmesiyle ili\u015fkilendirilmi\u015ftir [35].<\/p>\n<p>Zeytinya\u011f\u0131 polifenolleri, anti-inflamatuvar etkilerinin yan\u0131 s\u0131ra ba\u011f\u0131\u015f\u0131kl\u0131k sistemini d\u00fczenleyici \u00f6zelliklere de sahiptir. Bu \u00f6zellikler, IBH gibi kronik inflamasyonla ili\u015fkili hastal\u0131klar\u0131n yan\u0131 s\u0131ra multiple skleroz, sedef hastal\u0131\u011f\u0131, romatoid artrit ve sistemik lupus eritematozus gibi di\u011fer imm\u00fcn hastal\u0131klar\u0131n etkilerini azaltabilir [83].<\/p>\n<p>Otoimm\u00fcn ve inflamatuvar tepkilerde ana rol oynayan h\u00fccreler, T lenfositleri ve antijen sunan h\u00fccrelerdir (B h\u00fccreleri, monosit\/makrofajlar ve dendritik h\u00fccreler) [84]. \u0130nflamatuvar sitokinlerin (\u00f6rn. TNF-alfa, IL-8, IL-10, IL-6, IL-17) y\u00fckselmesi ve ba\u011f\u0131\u015f\u0131kl\u0131k h\u00fccrelerinin aktivasyonu IBH\u2019nin geli\u015fimi ve ilerlemesinde \u00f6nemli bir rol oynar. Bu do\u011frultuda, diyetle al\u0131nan zeytinya\u011f\u0131 polifenolleri, anti-inflamatuvar etkileri sayesinde IBH\u2019nin klinik ve patolojik seyrini de\u011fi\u015ftirebilir [85,86]<\/p>\n<h3><strong>Zeytinya\u011f\u0131nda Bulunan Bile\u015fikler ve Kolorektal Kanser \u00dczerindeki Etkileri<\/strong><\/h3>\n<ol>\n<li><strong>Apigenin<\/strong><br \/>\nApigenin, zeytinya\u011f\u0131ndaki flavonoid alt s\u0131n\u0131f\u0131na ait bir bile\u015fiktir ve y\u00fczy\u0131llard\u0131r geleneksel \u00c7in t\u0131bb\u0131nda kullan\u0131lmaktad\u0131r. Kolorektal, karaci\u011fer, meme, akci\u011fer ve prostat kanserlerinde anti-t\u00fcm\u00f6r etkiler g\u00f6sterdi\u011fi, d\u00fc\u015f\u00fck toksisiteye ve mutajenik aktiviteye sahip olmad\u0131\u011f\u0131 belirtilmi\u015ftir. Apigenin, JAK\/STAT, PI3K\/AKT, NF-\u03baB, MAPK\/ERK ve Wnt\/\u03b2-katenin gibi sinyal yollar\u0131n\u0131 mod\u00fcle ederek, doz ba\u011f\u0131ml\u0131 olarak h\u00fccre \u00e7o\u011falmas\u0131n\u0131, g\u00f6\u00e7\u00fcn\u00fc ve invazyonunu engellemi\u015ftir. Ayr\u0131ca, apigenin\u2019in Bcl-2 protein ailesini bloke ederek CRC h\u00fccrelerinde apoptozu art\u0131rd\u0131\u011f\u0131 g\u00f6zlemlenmi\u015ftir. Bu \u00f6zellikler, apigenin\u2019in diyet takviyesi veya kemoterap\u00f6tik ila\u00e7larla birlikte kullan\u0131m\u0131n\u0131 \u00f6nermektedir.<\/li>\n<li><strong>Luteolin<\/strong><br \/>\nLuteolin, zeytinya\u011f\u0131nda glikozitlenmi\u015f formda bulunan do\u011fal bir flavonoiddir. Ba\u011f\u0131rsakta emilim s\u0131ras\u0131nda serbest luteoline d\u00f6n\u00fc\u015ft\u00fcr\u00fcl\u00fcr. Luteolin, inflamasyon, oksidasyon ve kanserler \u00fczerinde faydal\u0131 etkiler g\u00f6stermi\u015ftir. Kolorektal kanser h\u00fccrelerinde G2\/M h\u00fccre d\u00f6ng\u00fcs\u00fc durmas\u0131na neden olmu\u015f ve apoptozu tetiklemi\u015ftir. Ayr\u0131ca, Nrf2\/ARE yolunu epigenetik olarak aktive ederek CRC\u2019ye d\u00f6n\u00fc\u015f\u00fcm\u00fc engellemi\u015ftir.<\/li>\n<li><strong>Maslinik Asit (MA)<\/strong><br \/>\nZeytinlerin mumsu kabu\u011funda y\u00fcksek seviyelerde bulunan maslinik asit, inflamasyonu mod\u00fcle ederek kronik inflamatuvar s\u00fcre\u00e7lerin \u00f6nlenmesine katk\u0131 sa\u011flayabilir. MA\u2019n\u0131n hem mitokondri kaynakl\u0131 (intrinsik) hem de h\u00fccre d\u0131\u015f\u0131 (ekstrinsik) apoptoz yollar\u0131n\u0131 aktive ederek kolon kanseri h\u00fccrelerinde apoptozu ind\u00fckledi\u011fi g\u00f6sterilmi\u015ftir. Bu \u00f6zellikleri, MA\u2019y\u0131 kemoterap\u00f6tik veya kemopreventif bir ajan olarak \u00f6ne \u00e7\u0131karmaktad\u0131r.<\/li>\n<li><strong>\u03b2-Sitosterol<\/strong><br \/>\nZeytinya\u011f\u0131nda bulunan bir fitosterol olan \u03b2-sitosterol, kolon kanseri h\u00fccrelerinin b\u00fcy\u00fcmesini doz ba\u011f\u0131ml\u0131 olarak inhibe etmi\u015f, reaktif oksijen t\u00fcrlerini (ROS) temizleyerek apoptoza neden olmu\u015ftur.
